Our goal was to investigate the role of the endocrine disruptor bisphenol A (BPA), and 17 β-estradiol (E2) in the regulation of endothelin-1 (ET-1) mRNA expression and ET-1 release. \n Methods: Cultured human umbilical vein endothelial cells (HUVECs) were incubated ± BPA (0.01-100 μ mol/L) and E₂ (0.001-10 μ mol/L) for 48h. We determined cell viability (measured using a cell counting kit), ET-1 release (measured by EIA) and preproendothelin-1 ( ppET-1) mRNA levels (evaluated by RT-PCR) after 0h, 24h and 48h.\nResults: Cell viabillity decreased to 89% (p<0.05) with 10 μ mol/L E₂ and to 78% (p<0.05) with 100 μ mol/L BPA.Endothelin release and ppET-1 mRNA were measured using lower, non-toxic E₂ and BPA doses. ET-1 release decreased to 87% and 84% with 10 and 100 nmol/L E₂ and to72% and 65% with 1 and 10 μ mol/L BPA (p<0.05 for all). E₂ (100 nmol/L) decreased ppET-1 mRNA leaves to 75% (p<0.05) and 85% after 24h and 48h, respectively. BPA (10μmol/L) decreased ppET-1 mRNA levels to 61% (p<0.05) and 80% after 24h and 48h, respectively.\nConclusion: E₂ and BPA directly attenuate ET-1 gene expression and decrease ET-1 release.
